Resequencing approaches

• High SNP density (1/60, maize)

  • Direct sequencing of amplicons from low copy number regions in the genome

• Low SNP density (soy: 1/350, human 1/1000)

  • Pre-screening of amplicons may be necessary before sequencing
    • Detection of mismatches in heteroduplexes
    • DHPLC, Chemical or enzymatic cleavage
    • Tradeoff depends on cost and SNP density
