Mapping was performed on a set of sixty-five recombinant inbreds derived by single-seed descent (F7) from a cross between the durum wheat cv Messapia (T. turgidum ssp. durum) and the accession MG4343 of T. turgidum ssp. dicoccoides.
Ninety-eight primer pairs of wheat microsatellites (WMS) developed for all chromosomes of the A- and B- genomes of hexaploid wheat were used for analysis. Fragment sizes were calculated by Fragment Manager 1.2 (Pharmacia) by comparison with internal size standards.
Markers were integrated into the existing RFLP map of durum wheat (see TAG-97-721, and Map_Data : T. turgidum, Messapia x dicoccoides), using the 'PLACE' and 'TRY' commands of the program MAPMAKER 2.0.
Eighty-one out of ninety-six (84.4%) microsatellites used for analysis detected polymorphisms, and eight-five loci amplified by seventy-nine primer pairs could be placed on the RFLP map of durum wheat.
