General Remarks
Amplicon: Group 1,2,4,5: 381bp. Group 3: 334bp
Amplification conditions: A touch-down PCR protocol was run as follows: initial denaturation at 94 degr. C for 30 seconds; annealing at 62 degr. C for 30 seconds, decreasing by 0.5 degr. C/cycle; extension at 72 degr. C for 45 seconds, followed by repeating these steps for eight cycles. After enrichment, the program continued for 29 cycles as follows: 94 degr. C for 30 seconds, 58 degr. C for 30 seconds and 72 degr. C for 45 seconds.

Comment
The majority of studies report that a single dominant gene determines red glume color. A few papers report two factors (Torrie, 1936; Vacenko, 1936; Miczynski, 1938). Red glume color in Swedish land cultivars is apparently associated with hairy glumes (Schmalz, 1958) suggesting, because Hg is located in chromosome 1A, that a red glume factor different from Rg1 is involved in the Swedish stocks. Nothing is known of the possible association of such a gene with Bg, another glume color gene on 1A. See (Zeven, 1983) for review.
