Amplification Conditions
Reaction mixture in 25 ul volume containing 50 mM KCl, 1.5 mM MgCl2, 10 mM Tris.HCl (pH 8.3), 0.2 mM each of dATP, dTTP, dGTP, and dCTP, 0.4 uM each primer, 2 units of Stoffel fragment, and 2.5 ul of template. PCR amplification performed at 95oC for 5 min, 30 cycles of 94oC for 55 s, 60oC for 1 s, and 72o C for 40 s, and final extension at 72oC for 5 min.
Specificity
Amplifies 5S rDNA spacer regions in monocots.
